چکیده

Farnesyltransferase inhibitors (FTIs) are a novel class of anticancer drugs that can reverse Ras transformation. One of the intriguing aspects of FTI biology is that continuous drug exposure is not necessary to maintain phenotypic reversion. For example, after a single exposure to FTIs, Ha-Ras-transformed fibroblasts revert to a flat and anchoragedependent phenotype that persists for many days after processed Ras has returned to pretreatment levels. In this study, we show that persistence of the reverted state is mediated by elevated expression of the collagen isoform a2(I), a suppressor of Ras transformation the transcription of which is repressed by activated Ras and derepressed by FTI treatment. To our knowledge, this is the first report identifying an FTI-regulated gene which is linked to phenotypic reversion. The finding that extracellular matrix alterations can influence the kinetics of reversion supports our assertion that Rho-regulated cell adhesion parameters are a crucial determinant of the cellular response to FTIs.

Modeling Breast Acini in Tissue Culture for Detection of Malignant Phenotype Reversion to Non-Malignant Phenotype

Backgrounds: Evidence is accumulating to support disruption of tissue architecture as a powerful event in tumor formation. For the past four decades, intensive cancer research with the premise of “cancer as a cell based-disease” focused on finding oncogenes or tumor suppressor genes. However, the role of the tissue architecture was neglected. Three dimensional (3D) cell cultures which can recap...

Transcriptional inhibition of Ha-ras in interferon-induced revertants of ras-transformed mouse cells.

Elevation in Ha-ras expression, due to transcriptional activation or gene amplification, is associated with oncogenic transformation of NIH 3T3 cells. We have previously shown that murine interferon (IFN)-alpha/beta induced phenotypic reversion of NIH 3T3 cells transformed by long terminal repeat (LTR)-activated c-Ha-ras. The revertants produced decreased amounts of ras-encoded Mr 21,000 protei...

Time dependent of epigenetic effect of disulfiram on tumor suppressor gene of RASSF1A in Hela cancer cell line

Introduction: Cervical cancer is the third most common tumor among women. Surgery, radiotherapy, and chemotherapy are common treatments, however high stage tumors have frequently poor prognosis. Nowadays, the epigenetic reversion introduced as an efficient strategy of treatment of cervical cancer. In the process, inhibitors of DNA methyltransferase (DNMT) induce re-expression of tumor suppresso...

Targeting the Ras signaling pathway: a rational, mechanism-based treatment for hematologic malignancies?

A series of alterations in the cellular genome affecting the expression or function of genes controlling cell growth and differentiation is considered to be the main cause of cancer. These mutational events include activation of oncogenes and inactivation of tumor suppressor genes. The elucidation of human cancer at the molecular level allows the design of rational, mechanism-based therapeutic ...
